Description

PA007754.h1Fs-b: Biotinylated Recombinant Anti-human CD4 Monoclonal Antibody (Clone: OKT4), Human IgG1 Fc Silent Kappa

Recombinant human IgG1 monoclonal antibody with site-directed biotinylation through the Avi tag at the C-terminus of the antibody heavy chain.

The recombinant OKT4 antibody binds to the human CD4 protein (cluster of differentiation 4), a glycoprotein and the co-receptor for the T-cell receptor (TCR). The CD4 protein is expressed on the surface of immune cells such as helper T cells, monocytes, macrophages, and dendritic cells.

1. Single amino acid substitution in the V3 domain of CD4 is responsible for OKT4 epitope deficiency

Kawamura, N., et al. Immunogenetics. 1991;34(3):208-10. PMID: 1716607

“The OKT4 antibody (anti-human CD4) was used to identify a deficiency in the CD4 epitope in patients with normal CD4+ T cell counts, as detected by other anti-CD4 monoclonal antibodies. This study linked a single amino acid substitution in the V3 domain of CD4 to the OKT4 epitope deficiency, relevant to understanding HIV pathogenesis as CD4 is the HIV receptor.”

2. Surface antigen changes occurring in short-term cultures of activated human T lymphocytes: analysis by flow cytometry

Boylston, A. W., et al. Cell Immunol. 1982 Jul 15;71(1):12-26. PMID: 6183008

“The OKT4 antibody (anti-human CD4) was used in flow cytometry to analyze surface antigen changes in activated T lymphocytes. The study observed a shift in the OKT4:OKT8 ratio from 60:40 to 40:60 in cultured T lymphoblasts, suggesting a possible phenotype change from OKT4+ to OKT8+ cells, while OKT4 expression remained stable.”

3. Reversible alterations in immunoregulatory T cells in smoking. Analysis by monoclonal antibodies and flow cytometry

Ferson, M., et al. Chest. 1982 Nov;82(5):526-9. PMID: 6982152

“The OKT4 antibody (anti-human CD4) was used in flow cytometry to assess T-cell subsets in heavy smokers, revealing a decreased percentage of OKT4+ cells and a reduced OKT4+/OKT8+ ratio. These changes normalized six weeks after smoking cessation, indicating reversible alterations in immunoregulatory T cells linked to smoking.”

4. Quantitative immunofluorescent assay of full-length, recombinant CD4 in solution and mapping of its epitopes

Lyding, J., et al. J Fluoresc. 1991 Jun;1(2):141-6. PMID: 24242964

“The OKT4 antibody (anti-human CD4) was used in a quantitative flow cytometry assay with Simply Cellular microbeads to measure recombinant CD4 antigen levels. The assay, using standard conditions for OKT4 binding and detection, enabled epitope mapping and determination of CD4 oligomerization levels in solution.”

5. Asbestos exposure correlates with alterations in circulating T cell subsets

de Shazo, R. D., et al. Am J Med Sci. 1985 Jan;289(1):1-5. PMID: 3929128

“The OKT4 antibody (anti-human CD4) was used in flow cytometry to assess alterations in circulating T-cell subsets in asbestos-exposed individuals. The study identified changes in CD4+ T-cell populations, suggesting microbial metabolites like ascorbate influence CD4+ T-cell function, with implications for Th17-mediated conditions like Crohn’s disease.”
