PA005171: Recombinant Mouse IgG2c Isotype Control Antibody (Clone: 12B9), In Vivo Grade

The mouse IgG2c antibody is produced in certain inbred strains such as C57BL/6, C57BL/10, SJL, and NOD, which does not express the mouse IgG2a antibody, and vice versa. If one uses any of the mouse strains for animal research, it is better to use the IgG2c antibodies rather than the IgG2a antibodies. There are more and more IgG antibody formats with Fc engineered. The format with the Fc silenced, Fc silent, or Fcs with silenced effector function is the most popular. Thus, the wild type mouse IgG2c isotype control and the Fc silenced mouse IgG2c isotype control mutants should be used in such an application.

Background

Mouse IgG antibodies are classified to four subclasses mainly based on the isotype of the heavy chain (IgG1, IgG2a or IgG2c, IgG2b, and IgG3 for the heavy chain; kappa and lamda for the light chain), of which IgG1 and kappa are the most abundantnamed in serum. IgG antibodies are large molecules of about 150 kDa composed of four peptide chains, two identical heavy chains of about 50 kDa and two identical light chains of about 25 kDa. The four peptides are arranged in a Y-shape tetramer by disulfide bonds formed between the two heavy chains and between a heavy chain and a light chain.

“The experimental design of the four in vivo experiments reported in this study is shown in … In addition to the sham and LPT treatments, the mice were treated with anti-mouse programmed cell death protein 1 (PD1) antibody (#PA007163.m2cLA, Syd Labs) or an isotype control antibody (#PA007141, Syd Labs). Four treatments of 200 μg of antibody diluted in 100 μL of PBS (2 mg/mL) were administered intraperitoneally between days 9 and 21 of the experiment. The anti-PD1 dosing regimen of 200 ug per injection was selected based on established murine immunotherapy protocols rather than allometric scaling from human doses, consistent with prior studies demonstrating biological efficacy at this dose [21, 22]. Mice were divided into four treatment groups: group 1 received sham treatment and isotype control antibodies; group 2 received LPT and isotype control antibodies; group 3 received sham treatment and anti-PD1 antibodies; and group 4 received LPT treatment and anti-PD1 antibodies. The cancer cells were injected on experimental day 0, treatments started on day 8, ended on day 22, and the mice were euthanized with carbon dioxide inhalation and on day 25.”
