Biotinylated recombinant anti-human CD20 monoclonal antibody (Clone 2H7), engineered with a Human IgG1 Fc-silent domain to eliminate non-specific Fc receptor binding during flow cytometric analysis. Features site-directed biotinylation via a C-terminal Avi tag on the heavy chain, with variable regions derived from anti-human CD20 clone (2H7) and expressed in mammalian cells.

  • Conjugation & Purity: Biotinylated (Site-directed via C-terminal Avi-tag), > 95% by SDS-PAGE
  • Applications: Flow Cytometry (FC) via Streptavidin Conjugates, Cell Surface Staining, B Cell Depletion Monitoring
  • Alternative Formats: Unconjugated human IgG1 Fc-silent, mouse IgG1, and other recombinant versions of clone 2H7 as well as B9E9 and 1F5 clones are also available at Syd Labs and Ushelf.

Description

PA007746.h1Fs-b: Biotinylated Recombinant Anti-human CD20 Monoclonal Antibody (Clone: 2H7), Human IgG1 Fc Silent Kappa

Recombinant human IgG1 monoclonal antibody with site-directed biotinylation through the Avi tag at the C-terminus of the antibody heavy chain.

References of Anti-Human CD20 Monoclonal Antibody (Clone: 2H7):

1. IgG-Based Bispecific Anti-CD95 Antibodies for the Treatment of B Cell-Derived Malignancies and Autoimmune Diseases.

Hörner, S., et al. Cancers (Basel). 2022 Aug 12;14(16):3941. doi: 10.3390/cancers14163941. PMID: 36010934

“The study utilized hybridoma-derived antibodies 2H7, 4G7, and Apo-1 for quantification. The bispecific antibodies (bsAbs) combined the CD95 agonist Apo-1 with either the anti-CD20 antibody clone 2H7 or the anti-CD19 clone 4G7 in F(ab)2 and single-chain formats. The CD95xCD20 bsAb demonstrated remarkable potency with IC50 values as low as 25 pM, despite the relatively low affinity of the anti-CD20 mAb clone 2H7 as a single chain. Expression of CD20, CD19, and CD95 on various cell lines was quantified using flow cytometry with QIFIKIT calibration beads, and binding of bsAbs to Daudi and Jurkat cells was assessed via flow cytometry.”

2. Phenotypically distinct anti-insulin B cells repopulate pancreatic islets after anti-CD20 treatment in NOD mice.

Boldison, J., et al. Diabetologia. 2019 Sep;62(9):1669-1678. doi: 10.1007/s00125-019-04974-y. PMID: 31037349

“In this study, female VH125.hCD20/NOD mice (6-8 weeks old) were treated with either anti-hCD20 antibody (clone 2H7) or control IgG2b antibody (clone MPC-11). Over a 12-week observation period, 48% of control-treated mice developed diabetes compared to 15% of 2H7-treated mice. The study involved injecting groups of 6- to 8-week-old VH125.hCD20/NOD mice with 2H7 anti-CD20 or IgG isotype control, followed by analysis of spleen and pancreatic lymph node (PLN) tissue for anti-insulin B cells post-treatment.”
