“Despite the usefulness of mouse models for studying human infections, differences in human and murine DCs, for example in their capacity to produce IL-12p70 and IL-1β in response to pneumococcal infection (Littmann et al., 2009), highlight the need for more research on human systems to study the effect of this viral/bacterial co-infection. …A recent patient study also described elevated levels of IL-12p70, IL-6 and IL-15 as markers for severe clinical outcome after IAV infection (Bermejo-Martin et al., 2009). …We found that IAV infection of MDDC cultures significantly enhanced the induction of IL-12p70, as well as IL-6, following SP infection. …Neutralizing type I IFN antibodies abrogated the induction of the IL-12p35 subunit confirming that IAV-induction of type I IFNs was the main pathway by which IAV primed MDDCs for enhanced IL-12p70 production following SP infection. …We found significantly elevated levels of IL-12p70 and IL-6 in IAV-infected MDDCs exposed to SP, while infection with either pathogen alone induced low or undetectable levels of both cytokines (Fig. 1A and B).”
