1. Spatial relationships between markers for secretory and endosomal machinery in human cytomegalovirus-infected cells versus those in uninfected cells.
Das, S. et al.J Virol. 2011 Jun;85(12):5864-79. doi: 10.1128/JVI.00155-11. PMID: 21471245
“Transferrin receptor (TFRC) is a key marker for endosomal compartments. Our objective is to understand the process of HCMV virion assembly that takes place in the cytoplasm. Studies of cVAC biogenesis and operation will be facilitated by a better understanding of the composition and three-dimensional (3-D) arrangement of the various substructures that form the cVAC. As part of developing a more complete structural definition of the cVAC, in this work we have compared in uninfected and infected cells the 3-D locations, extents of colocalization, and intracellular volumes of markers for the ER, Golgi apparatus, early and recycling endosomes, multivesicular bodies, and lysosomes, as well as markers associated with the ESCRT III apparatus. Although it was originally thought that viral proteins occlude cellular proteins from the cVAC, work from our and other laboratories has demonstrated that the cVAC consists of components of the secretory apparatus arranged as concentric cylinders.”
2. Cytotoxic potential of ribonuclease and ribonuclease hybrid proteins.
Rybak, S. M. et al.J Biol Chem. 1991 Nov 5;266(31):21202-7. PMID: 1939162
“Therefore RNase behaves like a potent toxin when directed into a cell. To explore the cytotoxic potential of RNase toward mammalian cells, bovine pancreatic ribonuclease A was coupled via a disulfide bond to human transferrin or antibodies to the transferrin receptor. The RNase hybrid proteins were cytotoxic to K562 human erythroleukemia cells in vitro with an IC50 around 10^-7 M whereas greater than 10^-5 M native RNase was required to inhibit protein synthesis. Cytotoxicity requires both components of the conjugate since excess transferrin or ribonuclease inhibitors added to the medium protected the cells from the transferrin-RNase toxicity. After a dose-dependent lag period inactivation of protein synthesis by transferrin-RNase followed a first-order decay constant.”
