Description

PA007456: Recombinant Anti-Human CD20 Monoclonal Antibody (Clone: B9E9), Mouse IgG2a Kappa, In vivo Grade

The in vivo grade recombinant anti-human CD20 Mouse IgG2a Kappa monoclonal antibody was produced in mammalian cells. The anti-human CD20 antibody is a specialized monoclonal antibody designed to specifically target and bind to the CD20 protein, a transmembrane antigen expressed on the surface of mature B-lymphocytes (B-cells). CD20 plays a key role in B-cell development, activation, and proliferation, but is notably absent on plasma cells and early progenitor B-cells.

What is Anti-Human CD20 Antibody?

An anti-human CD20 antibody selectively recognizes the CD20 marker, making it an essential tool in immunology research and clinical therapeutics. Popular research clones include 2H7 (widely used for flow cytometry to detect surface CD20) and L26 (preferred for immunohistochemistry due to its recognition of cytoplasmic epitopes).

Research Applications of Anti-Human CD20 Antibody

In laboratory settings, anti-human CD20 antibodies are commonly employed in:

  • Flow cytometry for identifying and quantifying B-cells in blood or tissue samples.
  • Immunohistochemistry (IHC) to visualize CD20-positive cells in tissue sections.
  • Immune profiling, studying B-cell function, and monitoring immune responses.

These antibodies enable precise detection of B-cell populations without affecting plasma cells, which do not express CD20.

Therapeutic Uses and Mechanisms

Therapeutic anti-human CD20 antibodies, such as Rituximab (the pioneering chimeric monoclonal antibody), work by depleting CD20-positive B-cells through mechanisms including antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and direct induction of apoptosis. They are widely used to treat:

  • B-cell blood cancers like non-Hodgkin lymphoma and chronic lymphocytic leukemia.
  • Autoimmune diseases, including multiple sclerosis (MS) and rheumatoid arthritis.

“The anti-human CD20 antibody rituximab is one of the most popular antibody drugs in human medicine and has been used for treating B-cell type lymphoma and autoimmune diseases for more than two decades. …The anti-human CD20 antibody, rituximab (Chugai Pharmaceutical Co., Ltd., Tokyo, Japan) and anti-hCD20-Ga-hIgG1fut (InvivoGen, San Diego, CA), a biosimilar obinutuzumab, were used as a type I and II antibodies, respectively. …However, neither bovine or feline CD20 were detected by 4E1-7-B_f in overexpressed NRK cells, and human CD20 was also not detected by 4E1-7-B_f in Ramos cells endogenously expressing human CD20. This result indicates that 4E1-7-B_f is specific to canine CD20. …Conversely, anti-human CD20 antibodies, rituximab and obinutuzumab biosimilar bound to canine CD20 overexpressed in NRK cells, although the binding intensity was lower than that of 4E1-7-B_f (KD value, 17.42 ± 1.76 × 10−9 M vs 4.2 ± 2.6 × 10−9 M). …Amino acids in red in dog CD20 represent amino acids substituted into human CD20, and M1 to M5 indicate that only that portion of canine CD20 was replaced by human CD20 amino acids.”

“In our previous study, we established an anti-human CD20 mAb (C20Mab-60) by using a Cell-Based Immunization and Screening method. …Therefore, the development of anti-CD20 antibody and the understanding of its molecular actions are thought to be important. …To clarify further characteristics of C20Mab-60, we performed epitope mapping using enzyme-linked immunosorbent assay (ELISA). …Previously, we developed an anti-human CD20 monoclonal antibody, C20Mab-60 (IgG2a, kappa), using the Cell-Based Immunization and Screening (CBIS). …CD20 is a glycosylated transmembrane protein and is expressed on normal B cells and B cell malignancies.”

3、Profiling the impact of anti-human CD20 monoclonal antibodies on lymphocyte B cell subsets and their precursors in the bone marrow and in lymphoid tissues in an immunocompromised mouse engrafted with human cells.

“The humanized model enabled us to study the depletion and replenishment of human B cells in various tissues using antibodies designed to specifically recognize human CD20 epitopes, that is hard to be performed in patients. …When we evaluated the repletion of B cells, OFA-treated mice showed preserved pre-B cells distribution in the bone marrow, as well as a recovery of immature B cells, followed by a progressive recovery of B cell distribution in the spleen and a partial recovery in the liver. …Seven days after the last injection a robust (>90 %) decrease of circulating human CD20+ B lymphocytes was observed in both OFA- and OCRE-treated mice. …A partial replenishment of B lymphocytes was detectable in blood 36 days from the last injection in OFA-treated mice, while no B lymphocytes could be detected in OCRE-treated mice up to 65 days post injection. …B lymphocytes are part of the adaptive immune arm and are involved in the production of antibodies.”

4、18F-labeled anti-human CD20 cys-diabody for same-day immunoPET in a model of aggressive B cell lymphoma in human CD20 transgenic mice.

“Both tracers were used for immunoPET imaging of the B cell compartment in human CD20 transgenic mice (hCD20TM). …The GAcDb was successfully 18F-radiolabeled using two different conjugation methods resulting in similar specific activities and without impairing immunoreactivity. …Fast blood clearance results in high contrast PET images as early as 1 h post injection enabling same-day imaging. …Metabolic imaging using [18F]FDG is the current standard for clinical PET; however, some malignancies (e.g., indolent lymphomas) show low avidity for FDG. …Both tracers ([18F]FB-GAcDb and [18F]FBEM-GAcDb) specifically target human CD20-expressing B cells in transgenic mice.”

5、B Cell Development in the Bone Marrow Is Regulated by Homeostatic Feedback Exerted by Mature B Cells.
Shahaf, G., et al. Front Immunol. 2016 Mar 22;7:77. doi: 10.3389/fimmu.2016.00077. PMID: 27047488
“To specifically address this question, we used mice transgenic for human CD20, where effective depletion of B lineage cells is obtained upon administration of mouse anti-human CD20 antibodies, in the absence of any effect on other cell lineages and/or tissues. …Compared to control mice, B cell-depleted mice exhibited a higher proliferation rate in the pro-/pre-B compartment, and higher cell death and lower differentiation in the immature B cell compartment. …In order to address this question, we have used the human CD20Tg mouse model, where peripheral B cells can be specifically eliminated using anti-hCD20 antibodies with no residual effect on other cell populations (25), in a study combining BrdU labeling over time and mathematical modeling. …For these experiments, we used Balb/c mice that are normal or transgenic for human CD20 (hCD20 Tg) (25) at 3–4 months of age. …Depletion was confirmed by flow cytometry analysis of peripheral blood cells.”
