The in vivo grade recombinant anti-HEL / HEL VHH bispecific antibody (mouse IgG2c-LALAPG kappa) was produced in mammalian cells. The second anti-HEL (HEL VHH) is a VHH or nanobody.

Background

A Mouse IgG2c bispecific isotype control antibody is a non-target-specific antibody that matches the host species, isotype, and kappa light chain of a mouse IgG2c bispecific antibody. It is used in various in vitro and in vivo assays like ELISA, Western Blot, Flow Cytometry, and animal model research to differentiate specific antibody binding from non-specific interactions.

The anti-HEL monoclonal antibody (hen egg-white lysozyme) in nature is suitable for use as an isotype-matched control antibody to estimate the degree of non-specific binding by an antigen-specific antibody. The HyHEL-10 / HEL D1.3-based recombinant isotype control and mutants (natural and engineered isotype controls) are useful for in vitro and in vivo studies from ELISA, Western blot (WB), to cell-based assay to animal model study. As an ideal recombinant mouse IgG2c bispecific isotype control antibody, the recombinant HyHEL-10 / HEL D1.3 mIgG2c kappa bispecific isotype control antibody has low or no specific binding to any mouse sample, and completely mouse variable regions and the mouse IgG2c kappa constant regions. The various formats of mouse IgG2c kappa bispecific isotype control antibodies and mutants can be used in ELISA, WB, the cell-based assay or animal model as the antibody negative control.

The immunoglobulin G1 (IgG1) kappa antibodies HyHEL-5 and HyHEL-10 interact with nonoverlapping epitopes on hen egg lysozyme (HEL); the HyHEL-5/HEL interface has two energetically and structurally important salt links, whereas the HyHEL-10/HEL interface involves predominantly hydrogen bonds and van der Waals interactions. The kinetics of association and dissociation of antibodies HyHEL-5 and HyHEL-10 with HEL under a variety of conditions were investigated in this study. The association of each antibody with HEL follows second-order kinetics. The association process is significantly diffusion-limited, as indicated by the viscosity dependence of the interaction of both antibodies with HEL, although detailed energetics suggest that the association process may be more complex. The association rate constant for the HyHEL-5/HEL system is within a factor of 2 of the modified Smoluchowski estimate for proteins of this size, whereas HyHEL-10 interacts with HEL with an association rate an order of magnitude lower.

The antigen binding fragment (Fab) of a monoclonal antibody (HyHEL-10) consists of variable domains (Fv) and constant domains (CL–CH1). Normal modes have been calculated from the three-dimensional structures of hen egg lysozyme (HEL) with Fab, those of HEL with Fv, and so on. In the HyHEL-10 system, the crystal structure of HEL–Fv (PDB code: 1C08), which is a complex of HEL and Fv, had no significant differences from that of HEL–Fab (PDB code: 3HFM), which is a complex of HEL and Fab. Figure 1 shows the schematic structure of HEL–HyHEL-10 (Padlan et al. 1989). Fab is composed of the light and the heavy chains (L and H).
