Background
Enterokinase (EK) is a serine proteinase that exhibits specificity for the sequence (Asp)4-Lys. It is the physiological activator of trypsinogen. The native EK enzyme has a heavy chain (115 kDa) covalently linked to a light chain or catalytic subunit (35 kDa). It has been well documented that the enterokinase light chain (EK) is catalytically effective (1), and thus most studies are performed using EK.

Unit Assay Condition: 20 mM Tris-HCl (pH 8.0 @ 25°C), 50 mM NaCl, 2 mM CaCl2, 25 ug of an MBP fusion protein test substrate and enzyme. Incubate at 23°C.
Formulation: a 0.2 um filtered solution in Tris, and NaCl.
Usage notes:
Suggested Reaction Conditions: Cleavage of an MPB-paramyosin-ΔSal fusion protein with an enterokinase site requires 1 IU per 50 ug.
Removal: Enterokinase will bind specifically to trypsin inhibitor agarose (e.g., Sigma T-0637).
