Background Information:

EPZ-5676 (Pinometostat) is a potent, selective and S-adenosyl
methionine (SAM) competitive inhibitor of DOT1L methyltransferase (Ki =
80 pM), displaying 37,000-fold selectivity over all other
methyltransferases tested. It inhibits H3K79 methylation and MLL-fusion
target gene expression, and potently and selectively kills acute
leukemia cell lines bearing MLL translocations. It is currently in
clinical trials.
