Celetrix electroporation technology enables high titer transient antibody production in CHO cells for R&D, toxicology studies, raw material manufacturing for IVD and research reagents, and beyond. Before introduction of Celetrix electroporation technology, most scientists use HEK293 cells to do protein and antibody transient production because the chemical transfection method for CHO cells is either low efficient or too costly if the ExpiCHO system is used, and the other CHO electroporation method for transient production is either low volume (100 ul) or too costly if the Maxcyte system is used. However, the industry prefers transient production using CHO cells if available, especially the volume in the range of 4 ml to a few liters.

Celetrix electroporation model SLT enables scalable transient protein production in CHO cells. The yields from 8 ml to 400 ml CHO transient production using 200 ul, 1 ml, 5 ml, and 10 ml electroporation are consistent because the principle is the same. The whole process is easy and convenient: CHO cells are spun down, resuspended with electroporation solution, and after 1-click and 2-second electroporation, transferred to CHO cell culturing media.

The yield of transient production in CHO cells using the cell electroporation model SLT is much higher than the other methods. The yield from transient gene expression (TGE) can easily reach 1 g/L for trastuzumab after 14-day cell culturing of the original 2x10E9 CHO cells in our hands.

In addition to DNA, the cell electroporation model SLT can be used for electroporation of RNA, siRNA, and proteins to mammalian cells including CHO cells.

Large scale antibody production with Celetrix Cell Electroporation Large-scale Model SLT
Model SLT (Single Large Tube) 10ml electroporator can electroporate 2x10E9CHO cells instantly for 400ml Fed-batch culture to reach g/L antibody expression level in 7 days. Manually processing 5 tubes for 2L Fed-batch culture would take only about 5 minutes.

Model SLT also supports 200ul and 1ml electroporation tubes for smaller scale testing. It can also be equipped with cell fusion accessories for hybridoma generation.
Model EX+ electroporator can process 1ml tubes for 2×108 CHO cells for 40ml Fed-batch culture to harvest about 40 mg antibody. It is also suitable for stable pool generation and high expression clone screening.
Compared to flow electroporation devices on the market, single large tubes run much faster and there is no efficiency loss suffered by flow electroporation. Both small scale and large scale Celetrix electroporators can reach 99% efficiency on CHO cells with small plasmids and over 80% efficiency with 11kb larger plasmids.

For even larger production scales much more than gram quantity, we recommend the stable pool method. Starting from 10ml electroporation, a stable expression pool can be generated in 10-14 days and it can be frozen and thawed for higher and more reproducible production.

Questions and Answers about CHO electroporation transfection transient production:

Questions: What is the problem with transient transfection to CHO cells?

Answers: Both chemical transfection and electroporation-based transfection methods have been tried to improve the efficiency and yield of protein transient production in CHO cells. So far, the efficiency of most reagents and kits is low for chemical transfection method for CHO cells; the exception is the ExpiCHO system. However, the cost of the ExpiCHO system is higher compared with the other reagents and kits in the market. The volume of most CHO electroporation systems for transient production is low (100 ul); the exceptions are the Maxcyte system and the SLT system. However, the cost of the Maxcyte system is higher compared with our CHO electroporation model SLT for transient production and the consumables. The SLT system is the most economic solution for higher titer antibody transient production in the scale from 4 ml to a few liters.

Questions: Can the electroporation model SLT be used for stable transfection in addition to transient transfection?

Answers: What results in the difference between transient transfection and stable transfection? Transient transfection exerts a temporary influence on cells because the external DNA fragments are not integrated to the host genome, whereas stable transfection leads to permanent genetic changes, by DNA integration into the host genome, that are usually passed on to future cell progeny. The plasmid vectors used make the major difference. The electroporation machine SLT can also be used to optimize CHO stable transfection.
