“Importantly, CXCR5 expression was only upregulated on Vγ1 γδ T cells after CFA immunization. …To investigate the involvement of different subtypes of γδ T cells on their ability to control the humoral immune response, 200 μg of anti-Vγ2 mAb (“anti-Vγ4”; UC3-10A6; BioXCell) or isotype control (BioXCell) were injected i.p. three days before either CFA or Alum immunization and three days before OVA booster dose. …Consistent with the increased levels of OVA-specific immunoglobulins following Vγ4 γδ T cell depletion, we found that Tfh cell frequency was higher in Vγ4 γδ T cell-depleted mice than in isotype control (IC)-treated mice immunized with CFA/OVA, but not after Alum/OVA immunization. …Importantly, TCRδ-/- mice transferred with CXCR5-/- γδ T cells had decreased Tfh cell frequencies and did not develop GC following CFA immunization as compared to mice transferred with WT γδ T cells. …Thus, CXCR5 expression on γδ T cells is induced by γδTCR activation.”
