Description
PA007461: Recombinant Anti-Human HLA-ABC Monoclonal Antibody (Clone: W6/32), Mouse IgG2a Kappa, In vivo Grade
The in vivo grade recombinant anti-human HLA-ABC Mouse IgG2a Kappa monoclonal antibody was produced in mammalian cells. The anti-human HLA-ABC antibody is a monoclonal or polyclonal antibody that targets conserved, non-polymorphic epitopes shared across human Major Histocompatibility Complex (MHC) Class I molecules (HLA-A, HLA-B, and HLA-C), which are expressed on the surface of nearly all nucleated cells. This antibody recognizes common regions of the HLA-ABC complex and is widely employed in immunological research for techniques such as flow cytometry, Western blotting, and immunohistochemistry to detect cell surface markers, investigate immune responses, and study transplantation-related mechanisms. HLA-ABC proteins play a critical role in immune surveillance and cell recognition, and the anti-human HLA-ABC antibody—particularly well-established clones like W6/32—has become a reliable and broadly used tool in the field due to its pan-specific reactivi

1、HLA class I antibodies trigger increased adherence of monocytes to endothelial cells by eliciting an increase in endothelial P-selectin and, depending on subclass, by engaging FcγRs.

“We investigated the mechanisms underlying monocyte recruitment by HLA class I (HLA I) Ab–activated endothelium. …We used a panel of murine mAbs of different subclasses to crosslink HLA I on human aortic, venous, and microvascular endothelial cells and measured the binding of human monocytic cell lines and peripheral blood monocytes. …Accordingly, HLA I mIgG2a but not mIgG1 treatment of endothelial cells significantly augmented recruitment, predominantly through FcγRI, and, to a lesser extent, FcγRIIa. …We confirmed these observations using human HLA allele-specific mAbs and IgG purified from transplant patient sera. …Importantly, the subclass of donor-specific Ab may influence its pathogenesis.”
2、Blockade of p-selectin is sufficient to reduce MHC I antibody-elicited monocyte recruitment in vitro and in vivo.
Hong, L., et al. Am J Transplant. 2013 Feb;13(2):299-311. doi: 10.1111/ajt.12016. PMID: 23279566
“Endothelial vesicles known as Weibel-Palade bodies (WPb) are mobilized after HLA I crosslinking, leading to adherence of the immature neutrophilic cell line HL60. …In addition, macrophage infiltration frequently accompanies antibody-mediated rejection (AMR), and correlates with donor specific HLA antibodies and poor graft outcome. …It is incompletely understood how HLA I antibodies may promote monocyte recruitment into the allograft. …This study demonstrates that selectin blockade prevents monocyte adherence in vitro and intragraft macrophage accumulation in vivo in response to MHC I antibodies. …The use of the human aortic tissue for the research described herein was approved by the OneLegacy Biomedical Review Board under the agreement #RS-02-10-2 and UCLA MTA2009-561.”
