Background of Recombinant Anti-Biotin Monoclonal Antibody

Recombinant mouse anti-biotin antibodies recognize biotin (vitamin B7) attached to proteins, peptides, nucleic acids, or solid matrices, and can be offered in fluorophore and enzyme-conjugated versions.

The anti-biotin monoclonal antibody can be used as a second-step reagent in various applications for detecting biotinylated targets. The biotin-streptavidin system is widely used in immunoassay signal amplification techniques because streptavidin binds biotin with extremely high affinity.

Because biotin is naturally present in a large variety of biological samples, it may affect the results of biotin-based assays, such as reduced sensitivity, false negatives or false positives. Streptavidin/biotin blocking kit, avidin/biotin blocking system, or biotin blockers are used to block signals from endogenous avidin, biotin and biotin-binding proteins in tissues when used with biotin-based IHC detection.

Sandwich ELISA for SEM derivatives was developed using an anti-SEM-NBA antibody and horseradish peroxidase-labeled avidin or anti-biotin antibody as a secondary conjugate. Anti-biotin antibody-HRP and avidin-HRP were purchased from Sigma-Aldrich (St Louis, MO, USA), biotin (S)-sulfoxide and lidimycin were purchased from TopScience (Shanghai, China), and 96-well ELISA plates were purchased from Corning (USA). The advantageous distance and optimal distance between epitopes under these two modes were also calculated. The failure to develop sandwich ELISA for small molecules is not directly caused by their molecular weight but largely by the steric hindrance caused by the small distance between epitopes. The obtained optimal EC50 values were 11.2 pg/mL (double-antibody sandwich with the epitope distance ≥16 Å) and 7.3 pg/mL (antibody-avidin sandwich with the epitope distance ≥17 Å).

Development of a novel flow cytometry method for detecting pneumococcal-specific B cells

PS was chemically biotinylated and mounted on anti-biotin beads, and labeled with phycoerythrin (PE)-conjugated anti-biotin antibody to form a PS-multimer used for cell staining. bPS-beads were washed once and then resuspended in 100 μl of washing buffer before labeling with anti-biotin-PE. Excess anti-biotin-PE was removed by washing the labeled bPS-beads three times. The ratio of bPS-beads to anti-biotin-PE was tested in a stepwise manner at v/v ratios of: 50:0.5, 50:1, 50:2, 50:4, and 50:8 for each pneumococcal serotype. A negative control tube containing anti-biotin-PE but no bPS monomer was used for each blood sample.

Receptors on endothelial and epithelial cells often recognize molecules that are expressed by fungi, and only a limited number of these receptors have been identified to date. Here, we describe a method for identifying novel host cell receptors for fungi that uses intact organisms to precipitate biotin-labelled host cell membrane proteins, which are then detected by immunoblotting with an anti-biotin antibody. Presented here is the method to use for identification of membrane proteins that bind to C. albicans.

When the complex reached the nitrocellulose membrane, the test line, coated with the immobilized anti-Biotin capture antibody, was captured with the Biotin tag on the other end of the amplicon. To adapt the Basic Kit for LFD, primers were modified with a 5′-FITC label and a 5′-Biotin tag, enabling amplicon capture and visualization on the dipstick via anti-FITC and anti-Biotin antibodies. The test was generated by specific anti-FITC and anti-Biotin capture antibodies that are compatible with the TiLV-specific primers tagged with FITC and Biotin. In this conjugate pad, where the gold nanoparticles (AuNPs) were coated, the AuNPs-anti-FITC probe was applied to bind with the FITC tag on each amplicon via antibody–antigen interactions, resulting in AuNPs-anti-FITC–DNA complexes that continued to migrate along the strip. When the complex reached the nitrocellulose membrane, the test line, coated with the immobilized anti-Biotin capture antibody, was captured with the Biotin tag on the other end of the amplicon.
