Our Ad5-RGD and Ad5/35 adenovirus production services include: 1) construction of an Ad5-RGD and Ad5/35 adenovirus vector for expression or silencing of a gene; 2) generation of Ad5-RGD and Ad5/35 adenovirus; 3) amplification and purification of Ad5-RGD and Ad5/35 adenovirus.
Ad5-RGD was modified with the introduction of an Arg-Gly-Asp (RGD)-containing peptide in the HI loop of the Ad5 fiber knob domain. The RGD-fiber modified Ad-RGD viruses have been extensively used to deliver genes into many cells and tissues, including primary bronchial epithelium, smooth muscle, endothelium, and macrophage and T cells.
Ad35 virus, which belongs to Adenovirus subgroup B, uses CD46 as a receptor. The Ad5/35 virus containing chimeric type 5 and 35 fiber proteins can infect CAR-negative cell lines and various human cell lines, such as macrophage cells, more effectively than Ad5 virus.
With a strong safety record, adenoviruses and recombinant adenoviral vectors have been widely used for gene function studies and human gene therapy. Recombinant adenoviruses that can not replicate are attractive vectors for gene delivery for the following reasons:
- they infect many types of mammalian cells with high efficiency;
- they do not disrupt the genome of the host cell but remain in the nucleus as episomal DNA;
- they provide transient, high level gene expression;
- they are suitable for both in vitro and in vivo applications.

The Ad5-RGD or Ad5/35 adenovirus is generated in HEK 293 cells using the Ad5-RGD or Ad5/35 adenovirus construct provided by a client.

BS081C. Amplification and purification of Ad5-RGD and Ad5/35 adenovirus.
Ad5-RGD or Ad5/35 adenovirus vectors supplied by clients or newly constructed by Syd Labs are amplified to 1 or 3 x 10^12 viral particles. The Ad5-RGD or Ad5/35 adenoviruses are purified by centrifugation using two sequential cesium chloride gradients. The final products (1 or 3 ml, 1x10^12 viral particles/ml) are dialyzed, titered spectrophotometrically, and tested for sterility.
